Prof. Olesen will coordinate the contribution from Glostrup Universiy Hospital (KASGlostrup). His expertise includes headache classification, (genetic) epidemiological and functional twin studies, such as cortical spreading depression and attack provocation. For the Consortium, he will perform clinical studies on migraine patients together with University of Duisburg-Essen (Uni Essen) and University of Liège (Ulg).
Summary of relevant expertise
- Neurophysiology of migraine provocation. Substances such as nitric oxide, more specifically its a pro-drug glyceryl trinitrate, histamine and calcitonin gene-related peptide can induce genuine migraine attacks. Nitric oxide inhibition is effective treatment of spontaneous attacks. Mechanisms of action of these drugs need to be elucidated. There are indications that the increase of cyclic GMP that occurs after nitric oxide (NO) administration is involves, since the phosphodiesterase 5-inhibitor sildenafil also very effectively induces attacks in migraine sufferers.
- Epidemiology and classification of migraine.Diagnosis in migraine is difficult, mainly because biochemical markers are lacking. Therefore, headache classification is important. Contribution to migraine (genetic) epidemiology by collecting large cohorts of twins, and collection of familial hemiplegic migraine (FHM) families.
